Growth Hormone Releasing Peptide-2
GHRP-2 (Pralmorelin, KP-102) is a synthetic hexapeptide and potent GH secretagogue receptor (GHS-R1a) agonist. Developed in the 1980s from research into enkephalin analogs that release GH, it is among the most potent GHS-R1a agonists per milligram. Approved in Japan as a diagnostic agent (GHRELIN) for GH secretion testing. Its GH-releasing potency exceeds Ipamorelin's but is accompanied by greater cortisol and prolactin stimulation.
GHRP-2 is a GHS-R1a agonist on pituitary somatotrophs (primary GH-releasing action) and on hypothalamic GHRH neurons (secondary amplification via stimulating additional GHRH release). This dual-site activity produces GH pulses larger than hypothalamic-only stimulation. GHRP-2 also stimulates cortisol via ACTH release and prolactin via pituitary lactotrophs. Combined with a GHRH analog (CJC-1295, Sermorelin), the two pathways activate complementary intracellular cascades — cAMP/PKA (GHRH) + IP3/PKC (GHRP) — for synergistic GH amplification.
