Human Growth Hormone (Somatropin)
Human Growth Hormone (HGH, Somatropin) is a 191-amino-acid single-chain polypeptide hormone produced by somatotroph cells of the anterior pituitary. Recombinant HGH (rHGH) produced via E. coli or mammalian cell expression is structurally identical to pituitary-derived GH. Multiple FDA-approved formulations (Genotropin, Humatrope, Norditropin, etc.) exist for GH deficiency, paediatric short stature, HIV-associated wasting, Prader-Willi syndrome, and other approved indications.
GH acts on GH receptors (GHR) — a member of the cytokine receptor superfamily — through JAK2/STAT5 signalling. Hepatic GHR activation is the primary driver of IGF-1 production, which mediates most anabolic effects on bone and skeletal muscle. Direct GHR activation on adipocytes stimulates hormone-sensitive lipase (lipolysis) and inhibits lipoprotein lipase (anti-lipogenic). Counter-regulatory effects on insulin signalling (reduced glucose uptake) account for GH's diabetogenic potential at supraphysiological doses.
GH deficiency (both childhood and adult-onset), cachexia and wasting syndromes (HIV, cancer), short bowel syndrome, body composition research (lean mass and bone density), paediatric growth disorders, age-related GH decline (somatopause), and surgical/burn recovery. As a research compound it also serves as the reference standard against which GH secretagogues are benchmarked.
