Growth Hormone Releasing Peptide-6
GHRP-6 (His-D-Trp-Ala-Trp-D-Phe-Lys-NH2) is a synthetic hexapeptide and the original GHS-R1a agonist, developed by Bowers et al. in the 1980s from enkephalin structural research. It is the founding member of the GHRP family and remains a reference standard for GHS-R1a pharmacology. Alongside significant GH release, it produces pronounced appetite stimulation — the most marked of the GHRP class.
GHRP-6 activates GHS-R1a on pituitary somatotrophs (GH release) and on arcuate nucleus neurons (orexigenic appetite stimulation via NPY/AgRP pathway upregulation). The strong appetite stimulation is mediated through hypothalamic ghrelin receptors in a manner analogous to endogenous ghrelin, making GHRP-6 both a GH secretagogue and a potent stimulant of food intake. Cortisol and prolactin stimulation is intermediate between GHRP-2 and Ipamorelin.
GH secretagogue pharmacology, appetite and food intake research, cachexia and anorexia models (where appetite stimulation is desirable), ghrelin axis biology, body composition, and GH-axis stimulation protocols. Gastric protection studies have shown GHRP-6 (like BPC-157) has direct gastroprotective effects, independently of its GH-releasing activity.
