Modified GRF(1-29)
CJC-1295 without DAC (also known as Modified GRF 1-29 or Mod GRF 1-29) is a 29-amino-acid analog of GHRH(1-44) stabilised by four amino acid substitutions (positions 2, 8, 15, and 27) but without the albumin-binding Drug Affinity Complex. This gives a half-life of ~30 minutes — substantially longer than endogenous GHRH (minutes) but far shorter than CJC-1295 DAC (days), producing discrete GH pulses rather than sustained GH elevation.
Acts on GHRH receptors (GHRHr) on anterior pituitary somatotrophs to stimulate GH synthesis and release, then clears quickly — mimicking a single physiological GHRH pulse. This pulsatile pattern is considered more biomimetic than sustained GH elevation from DAC forms. It is typically administered in combination with a ghrelin mimetic (Ipamorelin, GHRP-2) — the two receptor pathways (cAMP/PKA via GHRHr + IP3/PKC via GHS-R1a) are complementary, producing synergistic GH pulses 2-5× larger than either alone.
Modified GRF 1-29 is the most commonly studied short-acting GHRH analog in combination GH secretagogue protocols. Research encompasses GH-axis stimulation, body composition, recovery, sleep-associated GH release, and age-related GH decline. Its pulsatile GH release profile may better preserve hypothalamic-pituitary feedback compared to sustained-release forms.
