GHRH Analog with Drug Affinity Complex
CJC-1295 with DAC is a modified analog of growth hormone-releasing hormone (GHRH/GRF(1-44)), the hypothalamic neuropeptide that stimulates pituitary GH secretion. The DAC (Drug Affinity Complex) modification attaches a maleimido propionic acid group to lysine at position 28, enabling covalent binding to cysteine-34 of circulating albumin. This extends half-life from minutes (endogenous GHRH) to approximately 6-8 days, permitting once-weekly dosing in research protocols.
CJC-1295 DAC acts at GHRH receptors on somatotroph cells of the anterior pituitary to stimulate GH synthesis and release in a pulsatile pattern. The sustained albumin-bound depot maintains GHRH receptor occupancy over days, producing prolonged elevation in both GH and downstream IGF-1 levels. Unlike exogenous GH administration, this stimulates physiological GH release from the pituitary, preserving some feedback regulation.
Research has examined CJC-1295 DAC for GH-axis support in age-related GH decline, body composition research (lean mass preservation, visceral fat reduction), post-operative recovery, and IGF-1 upregulation. It is frequently studied in combination with ghrelin mimetics (Ipamorelin, GHRP-2) as these peptides act on the complementary ghrelin receptor pathway to provide synergistic GH pulse amplification.
