Delta Sleep-Inducing Peptide
DSIP (Delta Sleep-Inducing Peptide) is a synthetic nonapeptide (Trp-Ala-Gly-Gly-Asp-Ala-Ser-Gly-Glu) first isolated in 1974 from the cerebral venous blood of sleeping rabbits by Schoenenberger et al. at the University of Basel. Despite the name, research has shown it modulates sleep architecture and neuroendocrine function broadly rather than simply inducing sleep, with particularly interesting effects on HPA axis regulation and antioxidant systems.
DSIP modulates the hypothalamic-pituitary-adrenal (HPA) axis, reducing ACTH and cortisol release โ an effect relevant to stress-related sleep disruption. It enhances slow-wave (delta) sleep while reducing sleep fragmentation. Antioxidant activity (reducing lipid peroxidation and protecting mitochondrial membranes) has been documented in multiple tissue types. DSIP also interacts with opioid receptor sensitivity, with antagonistic effects at low doses on opioid tolerance development.
Research areas include sleep architecture optimisation, HPA axis dysregulation, chronic stress, cancer-related pain and insomnia, opiate withdrawal symptom mitigation, and menopausal symptom management (Russian clinical studies). Animal longevity studies showed that chronic DSIP administration extended mean and maximum lifespan in mice with reduction in spontaneous tumour incidence.
