FOXO4-DRI
FOXO4-DRI is a D-retro-inverso (DRI) peptide designed to disrupt the FOXO4:p53 protein-protein interaction that keeps senescent cells alive. Published by Baar et al. in Cell (2017), it represents one of the first rationally designed senolytics — compounds that selectively eliminate senescent cells while sparing healthy cells. The DRI modification (D-amino acids in reverse sequence) confers protease resistance and membrane penetration.
In senescent cells, the transcription factor FOXO4 forms a complex with p53 at the nuclear envelope, sequestering p53 away from mitochondria and preventing it from triggering intrinsic apoptosis. Senescent cells thereby persist in a pro-survival state despite accumulating damage. FOXO4-DRI disrupts this FOXO4:p53 interaction, releasing p53 to translocate to mitochondria and trigger BAX/BAK-mediated apoptosis — specifically in senescent cells. Non-senescent cells, lacking this FOXO4-p53 survival complex, are largely unaffected.
The original Baar et al. mouse study demonstrated that FOXO4-DRI selectively eliminated p21-high/p16-high senescent cells in naturally aged, fast-aged (XPD mutant), and chemotherapy-treated mice — restoring physical fitness, renal function, and fur density. Subsequent research has examined FOXO4-DRI in pulmonary fibrosis, cancer-therapy-induced senescence, and liver fibrosis models.
