GSH Glutathione
γ-L-Glutamyl-L-Cysteinyl-Glycine
Glutathione (γ-L-glutamyl-L-cysteinyl-glycine, GSH) is the most abundant intracellular antioxidant tripeptide, present at 0.5-10 mM concentrations in virtually all mammalian cells. Synthesised from glutamate, cysteine, and glycine by glutamate-cysteine ligase (GCL) and glutathione synthetase (GS). GSH levels decline with age, oxidative stress, toxic exposures, and chronic disease — making replenishment a focus of both anti-ageing and hepatoprotective research.
GSH serves multiple overlapping roles: as the primary electron donor for glutathione peroxidase (GPx) enzymes — the main intracellular defence against hydrogen peroxide and lipid hydroperoxides; as the redox buffer (GSH/GSSG ratio) that maintains the reducing intracellular environment; as a substrate for glutathione S-transferase (GST) enzymes that conjugate and neutralise electrophilic toxins and reactive metabolites; and as a direct participant in the gamma-glutamyl cycle for amino acid transport. In melanocytes, GSH inhibits tyrosinase, reducing eumelanin synthesis.
Intravenous glutathione has been studied for liver protection (hepatitis, alcoholic liver disease, chemotherapy-induced hepatotoxicity), detoxification support (heavy metal chelation adjunct, xenobiotic clearance), immune function enhancement (GSH is essential for T-cell proliferation and NK cell activity), skin brightening (tyrosinase inhibition), mitochondrial protection, and neurodegenerative disease (Parkinson's disease — intravenous GSH trials showed symptomatic improvement in early studies).
