Human Chorionic Gonadotropin
Human Chorionic Gonadotropin (hCG) is a heterodimeric glycoprotein hormone composed of shared alpha (92 aa) and unique beta (145 aa) subunits, produced by the placental syncytiotrophoblast during pregnancy. Recombinant hCG (Ovitrelle) and highly-purified urinary hCG preparations are used extensively in reproductive medicine. In the research setting, hCG functions as a long-acting LH surrogate due to its shared receptor and ~24-hour half-life (vs LH's ~30 minutes).
hCG binds the luteinising hormone/choriogonadotropin receptor (LHCGR) with high affinity, activating cAMP/PKA signalling in target cells. In Leydig cells, LHCGR activation upregulates StAR protein (mitochondrial cholesterol transport) and CYP11A1 (cholesterol side-chain cleavage enzyme), driving testosterone biosynthesis. In ovarian granulosa cells, the LH-surge equivalent produced by hCG triggers ovulation and corpus luteum formation and progesterone secretion.
Testosterone production stimulation in hypogonadism research, prevention of testicular atrophy and maintenance of intratesticular testosterone during androgen administration protocols, fertility protocols (ovulation induction, luteal phase support, spermatogenesis stimulation in hypogonadotrophic hypogonadism), and LH-receptor pharmacology. As the longest-acting endogenous-equivalent gonadotropin, hCG is a valuable tool in HPG-axis research.
